The smash left the driver of the Toyota, a 22-year-old man from Morphett Vale, with non-life threatening injuries. He was taken to Flinders Medical Centre for treatment.
Police are asking anyone who saw the silver Ford sedan or white Toyota Hilux travelling on Flaxmill Road prior to the crash to come forward.
Flaxmill Road was closed to all traffic for about eight hours while the scene was investigated.
It has since reopened.
The number of lives lost on South Australian roads this year is now 58, compared with 52 at this time last year.

Two teenagers have been taken to hospital and three others have minor injuries after a car crashed and flipped in South Australia s Riverland early this morning.
Emergency services were called to Garrett Road in Glossop after the black Suzuki Vitara crashed into a Stobie pole and landed on its roof about 1:30am.
Two passengers in the back of the car a 15-year-old girl and a 16-year-old boy were seriously injured and flown to the Royal Adelaide Hospital.
