The collision happened on Highway 25 north of Highway 182, according to Starkville police.
A northbound vehicle traveled into the southbound lane and struck an oncoming vehicle head-on.
Medics transported a third person to the hospital.
Oktibbeha County Coroner Michael Hunt said the third person suffered non-life-threatening injuries.
Hunt later identified the two deceased victims [both drivers] as Tabitha Snyder, 24, of Canton, Georgia; and Bonifacio Perez Pantoja, 38, of Columbus.
