Fundamental mechanism discovered that fine-tunes gene expression & is disrupted in cancer

Credit: The Wistar Institute PHILADELPHIA and MELBOURNE, Australia (May. 17, 2021) A team of scientists from The Wistar Institute in Philadelphia and the Peter MacCallum Cancer Center in Melbourne, Australia, discovered a new checkpoint mechanism that fine-tunes gene transcription. As reported in a study published in Cell, a component of the Integrator protein complex tethers the protein phosphatase 2A (PP2A) to the site of transcription allowing it to stop the activity of the RNA polymerase II enzyme (RNAPII). Disruption of this mechanism leads to unrestricted gene transcription and is implicated in cancer. The study points to new viable opportunities for therapeutic intervention demonstrating the anti-cancer effect of a new combination treatment in preclinical models of solid and hematopoietic malignancies.

Date Time New cellular atlas maps out healthy and cancerous breast tissue WEHI researchers have documented the diversity of cells in the human breast, explaining the relationship between healthy breast cells and breast cancer cells. The diversity of cells in the human breast has been documented in a new ‘RNA atlas’. The research, which relied on expertise spanning from breast cancer biology through to bioinformatics, measured gene expression in single cells taken from healthy women and cancerous breast tissue, including tissue carrying a faulty BRCA1 gene. This enabled the researchers to create an ‘RNA atlas’ that details the different cells found in these tissues.

Small proteins called monobodies were used to freeze MLKL at different stages as it moved from a dormant to an activated state, a key process that enables an inflammatory form of cell death called necroptosis. The team were able to map how the three-dimensional structure of MLKL changed, revealing potential target sites that might be targets for drugs - a potential new approach to blocking necroptosis as a treatment for inflammatory diseases.
