A Spanish rescue ship sped to the vessel after it was spotted from the air. Three boats with as many as 300 migrants went missing after leaving Senegal for the Canary Islands two weeks ago.
A rescue ship is on its way to the area where the missing vessel is believed to have been seen amid continuing fears for the safety of hundreds of migrants.
Two other vessels carrying dozens of people who travelled from Senegal remain missing, aid group says.
Two similar boats carrying dozens more people are also said to be missing.
