Frontiers | The One That Got Away: How Macrophage-Derived IL-1β Escapes the Mycolactone-Dependent Sec61 Blockade in Buruli Ulcer

Buruli ulcer (BU), caused by Mycobacterium ulcerans, is a devastating necrotizing skin disease. Key to its pathogenesis is mycolactone, the exotoxin virulence factor that is both immunosuppressive and cytotoxic. The discovery that the essential Sec61 translocon is the major cellular target of mycolactone explains much of the disease pathology, including the immune blockade. Sec61 inhibition leads to a loss in production of nearly all cytokines from monocytes, macrophages, dendritic cells and T cells, as well as antigen presentation pathway proteins and costimulatory molecules. However, there h...

Frontiers | Tilapia Lake Virus-Induced Neuroinflammation in Zebrafish: Microglia Activation and Sickness Behavior

In mammals, the relationship between the immune system and behavior is widely studied. In fish, however, the knowledge concerning the brain immune response and behavioral changes during brain viral infection is very limited. To further investigate this subject, we used the model of tilapia lake virus (TiLV) infection of zebrafish (Danio rerio), which was previously developed in our laboratory. We demonstrated that TiLV persists in the brain of adult zebrafish for at least 90 days, even when the virus is not detectable in other peripheral organs. The virions were found in the whole brain. Durin...

Frontiers | Review: Insights on Current FDA-Approved Monoclonal Antibodies Against Ebola Virus Infection

1 Abstract The devastating Ebola outbreak in West Africa in 2013-2016 accelerated the progress of several medical countermeasures (MCMs) against Ebola virus disease (EVD). Several IPs were used during that outbreak, but no conclusion on efficacy could be drawn. Only the Randomized Controlled Trial (RCT) on ZMapp was promising but inconclusive. More recently, during the second largest Ebola outbreak in North Kivu and Ituri provinces, Democratic Republic of the Congo (DRC), four IPs (Remdesivir, Regeneron, ZMapp and MAb114) have been evaluated in a RCT, the Pamoja Tulinde Maisha (PALM) study. Tw...
