New research discovered a key protein that plays a role in DNA double-strand break repair. MSH2-MSH3, a mismatch repair protein, is recruited to DSB sites through the interaction with a chromatin remodeler SMARCAD1 and facilitates EXO1 recruitment for long-range DNA resection. This helps to promote homologous recombination over error-prone polymerase theta-mediated end-joining pathway to repair DSBs. The findings provide a deeper understanding of the mechanisms underlying DSB repair and may lead to new cancer treatments.
