FDA says Voyager can trek on as it removes trial hold for Huntington's gene therapy test

The FDA is in a forgiving mood: After clearing uniQure’s gene therapy to restart trials today, it’s handing the same reprieve to Voyager Therapeutics.
The biotech was slapped with a clinical hold on an IND for the program, VY-HTT01, because of issues with chemistry, manufacturing and controls (CMC). This marked a miserable time for the program, coming one year after Sanofi’s Genzyme unit pulled out of a collab for it.
But now the FDA has lifted its hold on the gene therapy candidate in Huntington’s disease (HD), and “confirmed that the company may proceed with its planned phase 1/2 clinical trial,” according to a statement from the company, which plots to start this year in early HD patients. ....

Flagship's Repertoire bags $189M to go after immune targets

Repertoire Immune Medicines has raised $189 million to advance anti-cancer T-cell therapies through the clinic while expanding its immune response drug discovery platform.
Flagship Pioneering created Repertoire by combining two of its portfolio companies, Cogen Immune Medicines and Torque Therapeutics. The VC shop oversaw the merger to bring together the immune decoding and immuno-oncology platforms that underpinned Cogen and Torque, respectively, and put former Bioverativ CEO John Cox in charge of the combined company.
Now, Repertoire has raised a series B round to support the next stage of its development. Repertoire framed the financing primarily in the context of what it will mean for the DECODE technologies it has developed to characterize the immune synapse. ....

Tenaya grabs $106M top-up to push heart disease gene therapies to the clinic

With another $106 million in the bank, heart-focused Tenaya Therapeutics is ready to talk targets. The series C funding will propel several preclinical programs toward the clinic, including a gene therapy for hypertrophic cardiomyopathy (HCM), as well as set up a site in the Bay Area to manufacture vectors to deliver its treatments.
The South San Francisco-based biotech is attacking heart disease through three different angles: gene therapy, cellular regeneration and precision medicine. It’s taking this multipronged approach so it can solve the different types of problems that can lead to heart disease and heart failure. ....

Sanofi, following Denali, pens gene therapy pact with Sirion Biotech

French Big Pharma Sanofi is teaming up with Germany’s Sirion Biotech for its viral-vector-based gene delivery tech.
The pair will work together on developing improved tissue-selective adeno-associated virus (AAV) vectors to tee up better gene therapy treatments. Targets have not yet been specifically mentioned, simply for “disorders affecting major human organs.” Financials of the deal were not made public.
This comes two years after Sirion penned a similar pact with Denali Therapeutics, with other partners including Acucela and Orchard Therapeutics.
Sanofi will work with Sirion and the German company’s partner at Heidelberg University Hospital, Dirk Grimm, to develop new and modified AAV capsids. The overall goal is to make safer, better gene therapies. ....

Precision's one-time gene editing treatment lowers cholesterol in monkeys for 3 years

Precision BioSciences was founded in 2006 to develop a genome editing technology that uses homing nucleases to make precise edits in DNA. Now, the company has preclinical evidence showing that the approach designed as a one-time treatment holds promise for treating familial hypercholesterolemia, high cholesterol that’s caused by abnormalities in the gene PCSK9.
A single dose of Precision’s treatment reduced the expression of PCSK9, resulting in a stable lowering of LDL cholesterol in nonhuman primates for three years, according to a study led by the University of Pennsylvania and published in the journal Molecular Therapy. The treatment reduced PCSK9 protein levels by 85% and LDL cholesterol levels by 56%. ....
