ECM Integrity Assay In order to test the integrity of each assay’s ECM, 100 µg/mL of Collagen Type I – FITC conjugate (Sigma-Aldrich) was coated onto wells of an Oris™ assay plate (without stoppers) and a Costar ® plate. These plates were then incubated overnight at 37 °C / 5% CO 2. After incubation, stoppers were inserted into the Oris™ assay plate while sterile PBS was added to both assay plates’ wells. At 1 hour, 24 hour, and 48 hour intervals post-insertion, stoppers were removed before fluorescence images were acquired using a Zeiss Axiovert inverted microscope. Scratches were made in the Costar ® plate at each time point. Images were acquired using the same settings employed for the Oris™ assay plate. ....
