Frontiers | Crystal structures of glycoprotein D of equine alphaherpesviruses reveal potential binding sites to the entry receptor MHC-I - Vimarsana News

Frontiers | Crystal structures of glycoprotein D of equine alphaherpesviruses reveal potential binding sites to the entry receptor MHC-I

Cell entry of most alphaherpesviruses is mediated by the binding of glycoprotein D (gD) to different cell surface receptors. Equine herpesvirus type 1 (EHV-1) and EHV-4 gDs interact with equine major histocompatibility complex I (MHC-I) to initiate entry into equine cells. We have characterized the gD-MHC-I interaction by solving the crystal structures of EHV-1 and EHV-4 gDs (gD1, gD4), performing protein-protein docking simulations, surface plasmon resonance (SPR) analysis, and biological assays. The structures of gD1 and gD4 revealed the existence of a common V-set immunoglobulin-like (IgV-l...

Frontiers | Stiffening of Circumferential F-Actin Bands Correlates With Regenerative Failure and May Act as a Biomechanical Brake in the Mammalian Inner Ear - Vimarsana News

Frontiers | Stiffening of Circumferential F-Actin Bands Correlates With Regenerative Failure and May Act as a Biomechanical Brake in the Mammalian Inner Ear

The loss of inner ear hair cells causes permanent hearing and balance deficits in humans and other mammals, but nonmammals recover after supporting cells (SCs) divide and replace hair cells. The proliferative capacity of mammalian SCs declines as exceptionally thick circumferential F-actin bands develop at their adherens junctions. We hypothesized that the reinforced junctions were limiting regenerative responses of mammalian SCs by impeding changes in cell shape and epithelial tension. Using micropipette aspiration and atomic force microscopy, we measured mechanical properties of utricles fro...

Difference Between Oris Cell Migration Assay and Scratch Assay - Vimarsana News

Difference Between Oris Cell Migration Assay and Scratch Assay

ECM Integrity Assay In order to test the integrity of each assay’s ECM, 100 µg/mL of Collagen Type I – FITC conjugate (Sigma-Aldrich) was coated onto wells of an Oris™ assay plate (without stoppers) and a Costar ® plate. These plates were then incubated overnight at 37 °C / 5% CO 2. After incubation, stoppers were inserted into the Oris™ assay plate while sterile PBS was added to both assay plates’ wells. At 1 hour, 24 hour, and 48 hour intervals post-insertion, stoppers were removed before fluorescence images were acquired using a Zeiss Axiovert inverted microscope. Scratches we...
