LncRNA FTX enhanced the angiogenesis in stroke by acting as a sponge of miR-342-3p to regulated the expression of SPI1 level
Significance Pancreatic ductal adenocarcinoma (PDAC) is the most common form of pancreatic cancer and is one of the most deadly cancer types, with a 5-y survival rate below 10%. One reason for this high mortality rate is that PDAC cells have an enhanced ability to survive and proliferate despite existing in nutrient-deprived environments. Understanding the metabolic rewiring that enables nutrient scavenging and rapid metabolic processing of PDAC cells can provide new strategies to develop effective treatment options for this intractable disease. In this study, convergent findings reveal that ...
Although targeting BRAF mutants with RAF inhibitors has achieved promising outcomes in cancer therapy, drug resistance remains a remarkable challenge, and underlying molecular mechanisms are not fully understood. Here, we characterized a previously unknown group of oncogenic BRAF mutants with in-frame insertions (LLRins506 or VLRins506) of αC-β4 loop. Using structure modeling and molecular dynamics simulation, we found that these insertions formed a large hydrophobic network that stabilizes R-spine and thus triggers the catalytic activity of BRAF. Furthermore, these insertions disrupted BRAF...
Abstract Duchenne muscular dystrophy (DMD) is a fatal muscle disease caused by the lack of dystrophin, which maintains muscle membrane integrity. We used an adenine base editor (ABE) to modify splice donor sites of the dystrophin gene, causing skipping of a common DMD deletion mutation of exon 51 (∆Ex51) in cardiomyocytes derived from human induced pluripotent stem cells, restoring dystrophin expression. Prime editing was also capable of reframing the dystrophin open reading frame in these cardiomyocytes. Intramuscular injection of ∆Ex51 mice with adeno-associated virus serotype-9 encodin...
